亚洲玉米螟中肠类钙黏蛋白Cry1A毒素结合区对Cry1Ac毒素杀虫活性的影响

Effect of Cadherin-like Protein Cry1A Binding Region in Ostrinia furnacalis on Insecticidal Activity of Cry1Ac

摘要: 通过原核表达及亲和层析获得亚洲玉米螟Ostrinia furnacalis中肠类钙黏蛋白Cry1A毒素结合区MPR及其在Cry1Ac抗性玉米螟中的突变体MPR-r2多肽片段。采用饲料混合法测定了MPR和MPR-r2及其分别与Cry1Ac毒素混合对亚洲玉米螟幼虫的杀虫效果。结果表明,MPR和MPR-r2多肽片段对亚洲玉米螟幼虫都有一定的杀虫作用;当MPR多肽片段与Cry1Ac毒素混合时,杀虫活性具有加性效应,而MPR-r2多肽片段与Cry1Ac毒素混合的杀虫效果则没有显著提高。

Abstract: The Cry1A binding region of cadherin-like protein of the Asian corn borer (ACB), Ostrinia furnacalis, MPR, as well as one of its mutants in Cry1Ac-selected strain of ACB, MPR-r2, were expressed in Escherichia coli as a soluble protein and purified on a Ni-NTA column with its His-tag. The toxicities of MPR and MPR-r2 to ACB were examined using diet incorporate bioassay. The purified MPR and MPR-r2 peptides showed low toxicity to ACB. In addition, an additive effect on the insecticidal activity against the ACB larvae was observed when MPR peptide was mixed with Cry1Ac toxin. However, ACB mortality was not significantly increased when purified MPR-r2 peptide was mixed with Cry1Ac toxin as compared with Cry1Ac used alone.
